Installation of Moisture Barrier + Underfloor Insulation

We live in a house built in 1970 and we're noticing an build-up of mould. We would like to install a moisture barrier under the house where we can. Our master bedroom is 600 off the ground, so instead of a moisture barrier, we would like to fill this with polystyrene insulation and have it completely enclosed with marine ply to stop the cold southerly wind. We're also open to any other ideas or solutions to prevent the build up of mould or increase the overall comfort of the property. Double Glazing is already in place.
