Locate source of musty smell 2

Our 1980 house had a downstairs toilet that gets a strange unpleasant musty smell, almost like stale cigarette smoke. Most noticable at night times and particularly after rain. Not sure if mould in wall framing or perhaps an insect colony, chemical reaction, or something exuding from floor slab. Would like source identified and ideally rectified
