Ants in roof space

We seem to have an ant nest in our roof, they are coming through our sarked wood ceiling and little dead flying ants also fall out a skylight. The sarked ceiling has no roof cavity, but most of the rest of the roof including the part with the skylight has a small cavity. (Skylight and area where they come through on sarked ceiling are really close together) We have tried spraying them, bunging up the gaps, but they keep coming! Not sure what to do or what solution is - help appreciated
